Try it on paper first. A hint, the key idea and the worked solution are here when you need them.
Three amino acids have no ionisable side chain. Serine has and . Leucine has and . Threonine has and . Find the isoelectric point of each.
The isoelectric point is the pH at which the average charge is zero. For a neutral side chain, . For other amino acids, average the two pK values on either side of the neutral form. The fraction of a group that is still protonated is for a group that is positive when protonated.
Have you tried it?
Work it out on paper first. It is fine to get stuck or make a mistake: that is how we learn. When you are ready, go through the solution one step at a time, and check each step against your own.
Plan. The neutral zwitterion is the main form between and , so pI is their mean.
Step 1. Serine. .
Step 2. Leucine. .
Step 3. Threonine. , which is .
Answer: pI of serine 5.68, of leucine 5.98 and of threonine 5.60.
Check. All three are close to 6, as for glycine. Amino acids with a neutral side chain have similar pI values.
